Results indicated that across informants (parents and children, boys and girls) gastric banding surgery and weight loss pills were perceived as less acceptable forms of treatment than diet, exercise, and family behavior therapy. Bmi-1 collaborates with H-Ras to promote mammary epithelial cell transformation, tumorigenesis, and metastasis. $49.99 The polycomb group protein Bmi-1 is a transcription repressor reported to regulate self-renewal of normal and cancer stem cells, and prevent cellular senescence through inhibition of cyclin-dependent kinase inhibitors pi6lnk4a and p19Arf. Originally discovered as a cooperating oncogene with c-Myc in a murine model of leukemia, overexpression of Bmi-1 has since been reported in several forms of cancer, including breast cancer. A direct or collaborative role of Bmi-1 in the pathogenesis of breast cancer would be critical in the development of novel biomarkers for diagnosis and treatment of this disease. We demonstrate that overexpression of Bmi-1 alone and in combination with H-Ras in normal mammary epithelial cells (MCF10A) markedly changes cellular morphology, increases proliferative indices, and decreases the apoptotic response to apoptosis-inducing agents in vitro. Our data also suggests that overexpression of Bmi-1 confers stem cell-like properties, including the formation of spheroid structures resembling mammospheres in culture and overexpression of stem cell markers. Xenograft experiments in immunodeficient (SCID) mice demonstrate that (1) Bmi-1 overexpression alone in MCF1OA cells cannot induce tumors; (2) H-ras overexpression induces development of tumors with vascular, smooth muscle, and mast cell components; whereas (3) the combination of Bmi-1 and H-Ras induces development of poorly-differentiated aggressive mammary neoplasms with greatly increased propensity for spontaneous distant metastasis to liver, spleen, and brain. Moreover, by using tail vein injection experiments in SCID mice as a model of direct hematogenous spread to the lungs, combination of Bmi-1 and H-ras overexpression in mammary epithelial cells results in fulminant pulmonary metastasis leading to clinical signs of weight loss and dyspnea, whereas injection of cells overexpressing H-ras or Bmi-1 alone does not result in this aggressive metastatic phenotype. Effect of mindfulness meditation and home-based resistance exercise on weight loss, weight loss behaviors, and psychosocial correlates in overweight adults. $49.99 Purpose. To examine the effect of mindfulness meditation and home-based resistance exercise when added to a standard behavioral intervention on weight loss, weight loss behaviors, and various psychosocial correlates in overweight adults across 6-months. Methods. Seventy-one subjects (BMI = 32.9+/-3.7 kg/m2; age = 45.1+/-8.3 years) participated in a 6-month weight loss intervention. Subjects were randomly assigned to one of three treatment groups: standard behavioral weight loss program (SBWL, n=24), SBWL plus resistance exercise (RT, n=23), or SBWL plus mindfulness training (MD, n=24). Participants were instructed to decrease energy intake to 1200–1500 kcal/d and dietary fat intake to 20–30% of total energy intake, increase physical activity to 300 min/wk, and attend weekly group meetings. Resistance training using resistance tubing and exercise balls was added to SBWL+RT. SBWL+MD consisted of mindfulness training using meditation, yoga, and mindful eating techniques. Body weight, process measures of weight loss (physical activity, energy intake, eating behavior inventory), and psychosocial correlates of weight loss (eating and physical activity self-efficacy, outcome expectations (benefits) and barriers to exercise, dietary restraint and disinhibition, and body image) were measured at 0, 3, and 6 months. Results. Weight significantly decreased in all groups at 6 months (p<0.05), but did not differ between groups (SBWL=-6.1+/-2.4 kg; SBWL+RT=-8.8+/-1.9 kg, SBWL+MD=-8.0+/-0.2 kg). Physical activity (833+/-439 kcal/wk) and weight loss eating behaviors increased (p<0.05) with no significant difference between groups. Physical activity and eating self-efficacy, dietary restraint, and most subscales of body image increased over the 6-month intervention, while dietary disinhibition, perceived hunger, and overall exercise barriers decreased significantly over time, with no difference between the groups. Effects of nondirective and directive support on weight loss in an e-counseling intervention. $49.99 Nondirective support (NS; collaborative, flexible) is positively associated with successful disease management, healthy lifestyles, quality of life and support satisfaction, whereas directive support (DS; prescriptive, protocol driven) has negative or no associations with these outcomes. However, DS is advantageous in acute, stressful situations and when an individual lacks skills to handle a challenge. This study manipulated these support types through e-mail exchanges and evaluated their effects on weight loss, physical activity, dietary behavior, engagement, alliance, and satisfaction in a 12-week e-counseling program. Overweight individuals (N = 104) were randomly assigned to one of three conditions: NS, DS, or minimal support (MS). All participants received two e-mails each week: (1) weight loss lesson and web link to report weight, daily calories, and exercise; (2) feedback graphs. Participants in the NS and DS conditions also received individualized weight loss support following protocols reflecting each type of support. Participants in the NS condition reported greater NS and a greater ratio of NS to DS than participants in the DS condition indicating participants were sensitive to the support manipulation. There was no effect for weight loss based on treatment condition. Secondary analyses revealed a gender-by-condition interaction. Females lost more weight in the DS condition (10.61 pounds) than in the NS (5.51 pounds) or MS (5.98 pounds) conditions (number of males insufficient for between-group analyses). Participants in the DS condition had greater changes in physical activity than participants in the NS and MS conditions. There were no group differences for dietary behavior and engagement. Those in NS reported greater goal collaboration than those in DS and greater goal collaboration, task orientation, and bond than those in MS.
